Investigation of the effects of ozone treatment in endotoxin induced shock model in rats

Abstract (EN)
Current treatment of sepsis aims for resolving complications as hypoperfusion and hypotension, and for controlling infection source. Despite these interventions, sepsis keeps being disease with high mortality rates. Intensive studies for definitive treatment of sepsis haven?t given promising results yet. In this study, firstly systolic blood pressures (SBP) and heart rates (HR) of rats measured with tail-cuff method. Then, saline and lipopolysaccaride (LPS) (10 mg/kg, i.p) applied to control and treatment groups, respectively. At the first hour, ozone applied to treatment groups at three different doses (0.1, 0.3, 1.0mg/kg, i.p.) and to ozone group (1.0 mg/kg, i.p). SBP and HR were measured at 1st, 2nd, 3rd and 4th hours. At the end of the experiment, animals were sacrified and levels of nitric oxide (NO), asymmetric dimethylarginine (ADMA) and total antioxidant capacity (TAC) were measured in plasma samples that were collected from animals. In control and ozone groups, before and after the treatment no significant difference observed in SBP and HR values. Descrease in SBP and increase in HR occured with LPS were statistically significant and ozone application did not reverse this changes. However reducement in TAC and increasement in NO levels occured with LPS reversed by ozone at 1 mg/kg and 0.1 mg/kg doses, respectively. Plasma ADMA levels did not show any difference between groups. Due to reversing the changes in NO and TAC levels in endotoxic shock model, results suggest that ozone might be useful to correct the biochemical changes in sepsis that clinical counterpart of this model.
