Biochemical investigation of the protective effect of dexpanthenol (provitamin B5) against renal damage caused by carbon tetrachloride (CCl4) in rats

Abstract (EN)

Aim: Many toxic substances found in nature and chemical drugs used in clinics cause serious damage to different organs of the body. Compared with other organs, the kidneys are more sensitive to toxic chemicals, causing kidney cell damage not to be fully understood. In our study, we aimed to investigate the protective effect of dexpanthenol against kidney damage in rats treated with carbon tetrachloride. Material and Method: 28 Spraque Dawley male rats were divided into four groups including control. Toxic effects of single dose of 0.5 ml / kg of carbon tetrachloride, and 500 mg / kg / day dexpanthenol as a protective was administered every day for two weeks intraperitoneally. At the end of the experiment, the thorax of rats was opened under ketamine/xylazine anesthesia and blood samples were taken by intacardiac punction. Kidneys were taken and stored in a -80oC deep freezer for biochemical and histopathological examination of the resulting oxidative damage. Results: A statistically significant difference was found between the groups according to the results of all biochemical analysis. It was observed that the control group had the highest GSH and TAS levels while CCl4 group had the highest MDA, TOS and OSI levels. Histopathologically, when the control group and CCl4 group were compared, it was observed that the tissue damage in CCl4 group increased statistically significantly. When the CCI4 group and CCl4+Dexpanthenol group were compared, it was found that the histopathological changes in this group were statistically significantly reduced. Conclusion: The results of the study show that dexpanthenol may be protective against oxidative damage caused by carbon tetrachloride in rat kidney tissue.
