Protective effect of proanthocyanidin against methotrexate induced injury in rat testis

Methotrexate, a drug used in chemotherapy spermatogenesis in male reproductive system, sperm quality, germ cells and results in damage to the blood-testis barrier is known to cause infertility in male subjects. The protective effect of the proanthosiyanid obtained from grape seeds on his damage in rat testis of methotrexate was investigated. In our research 26 Wistar Albino rats weighing 200-250 g divided four groups. We gave 2 cc/kg salin by gavage to the control group for 7 days. The proanthocyanidin groups received 200mg/kg proanthocyanidin by gavage for 7 days. The methotrexate group received single dose 20 mg/kg methotrexate by i.p at third day. The proanthocyanidin+methotrexate group received 200 mg/kg proanthocyanidin by gavage for 7 days and single dose of 20 mg/kg by i.p methotrexate only third day. At the eighth day all rats were sacrified. The testis tissue of all rats were taken. In addition to electron and light microscopic examinations, SOD, GPX and MDA analyses were done. In the seminiferous tubule epithelium in methotrexate-treated group a decrease in testicular histopathological findings such as thinning and spermatogonia cells. Electron microscopic findings also support these damages. At the same time significant increase in testicular tissue viewed MDA level, SOD and GPX levels also decreased significantly. In this study it was observed that tissue damage caused by methotrexate and prevent deterioration of the biochemical parameters of proanthocyanidine.
