Study of The Relationship Between Prolactin and Infertility by Measuring the expression Levels of Kiss1, Kiss 1 Receptor, Neurokinin, Neurokinin Receptor Genes in Rat Hypothalamus Cells Through Real Time PCR Prolactin hormone, which is secreted from the hypophysis and plays role in production of milk, has a polypeptide structure. Prolactin plays role in physiological situation such as metabolism and reproductivity. Increase in prolactin blood level leads to various disorders. The most important disorders of these are amenore/galactore and anovulation. Anovulation is a critical problem and gives rise to infertility. The ovulation can be controlled by hormones and biological substances called neuropeptide.It has been considered that kisspeptin and neurokinins amoung neuropeptides are involved in control of the ovulation. Kisspeptins, codded by Kiss1 gene separately expressed in neuronal populations of hypothalamus, are neuropeptide families. They play in roles timing of puberty and fertility. Furthermore, the other neuropeptide which has been thought effective in these situations is Neurokinin B (NKB). In recent neuroanatomic studies, it has been shown that NKB is synthesized in particularly hypotalamic neurons and arcuate nucleus which responses to sexual steroids. So it has been put forward that NKB affect sindirectly KISS1 synthesis due to presence of NKB in this region. It has been thought that NKB and KISS1 interact with hormones involved in puberty and reproductivity. We think that association between height of prolactin and infertility are relation of expression of kisspeptin and NKB. Therefore, in our study we investigated association between Kiss1, Kiss1 receptor, neurokinin and neurokinin receptor and infertiliy by increasing in blood prolactin level in rats due to high homology between human and rat. It has been shown that in rats which have high prolactin level, expression values of Kiss1, Kiss1 Recepror and neurokinin genes decreased accorging to control group in our data (p<0,001). As a result, we believe that neuropeptides mediate adverse effect of height of prolactin on reproducticiy and our study shed light of solution of infertiliy problems caused by height of prolactin.
