Investigation of biochemical parameters in rats after renal ischemia and hepcidin linked genes (BMP6, GDF-15, HJV)) which are responsible for the iron metabolism

Purpose: As well as its central role in regulation of iron homeostasis hepsidin alsoplays significant role in the body?s defense mechanism and inflammation. In order todetermine association of hepsidin with iron homeostasis, anemia and diseases thatcauses anemia was investigated on myocardial and liver ischemia/Reperfusion (I/R)models, but there has not been a study on the renal I/R model. In this study, the level ofserum biochemical markers [complete blood count, erythropoietin levels, reticulocytecount, iron (Fe), transferrin (TfR), solubl transferrin receptor (sTfR), hepcidin and IL-6]was evaluated after renal I/R. Also, in order to shed light on the diagnosis and treatmentof anemia, renal disease, and the other inflammatory disorders the expression levels ofhepsidin linked genes (GDF-15, BMP-6, HJV/HFE2) was investigated by using RTPCRtechnique. In addition, the histopatology of tissues were evaluated.Materials & Methods: A total of 20 Wistar albino rats were equally divided into 2groups. Group I: control group (n = 10), Group II = I/R group (60 minutes ischemia+ 48 hours of reperfusion) (n = 10). First group was control group and noting was doneand second group was experimental group therefore, in the left kidneys of these rats I/Rwas performed. For the expression studies right kidney of the experimental group wasused as a reference. Then, in the second group I/R generated parts of left renal arterieswas removed. Likewise right renal arteries of the all rats were removed. Furthermore,normal tissues of I/R generated kidneys were taken as a control.Findings-Results: As a result of renal dysfunction and I/R no significant associationwas found in terms of biochemical parameters hgb (p=0.593), RBC (p=0.842), iron (Fe)(p=0.364), Ret % (p=0.402), Tfr (p=0.849), sTfR (p=0.895), Epo (p=0.283), IL-6(p=0.111) and hepcidin (p=0.149). However, platelet (p=0.043) values were statisticallysignificant (p<0,05). Furthermore, expression levels of GDF-15 (p=0,126), BMP-6(p=0,420) and HJV/HFE2 (p=0,673) genes were not statistically significant. In addition,the evaluation of the general tissue histopathology was performed by usinghematoxylineosin (HE) and in terms of hypoxic-ischemic signs histopathology resultsshowed significant associations between control group and I/R group of ischemic andnon-ischemic kidney scoring. In addition, hypoxic-ischemic signs were also significantbetween I/R group of kidneys without ischemia and I/R group of kidneys with ischemia.
