Cellulase enzyme producing strain isolation and determination of the effect of medium conditions on enzyme activity

In this study, isolation of cellulase enzyme producing strains from soil and water resources was investigated. The cellulase enzyme activity of the isolated strains was determined by analyses performed on solid and liquid enzyme production media and the strain with the highest activity was selected. Selected strain having maximum cellulase activity was identified by 16S rRNA analysis as well as biochemical and morphological tests. It was determined that the selected strain showed maximum cellulase enzyme production at a temperature of 45 ° C and pH 6.0. In the different culture conditions used for enzyme production, it was determined that the best carbon source is starch, the best nitrogen source is yeast extract, and the K2HPO4 is the inducer component in enzyme production as a micronutrient. It was determined that the cellulase enzyme obtained has an optimum activity under the conditions of 7.5 of pH, 50 °C of temperature and 2% of NaCl concentration.
