Oxidative stress and krebs cycle enzyme levels in children with cerebral palsy

Cerebral palsy (SP); It is a non-progressive, permanent, movement-limiting, loss of motor function, posture and movement disorder as a result of an effect on a developing brain that occurs in the prenatal period, natal period or after the postnatal period. It is thought that deficiency in vitamin and nutrient intake, environmental factors and epileptic seizures may cause oxidative stress in children with CP. As a result of the researches, it was determined that the increase in oxidative stress has an effect on the Krebs cycle (TCA) enzyme activity levels. Our aim in this study is to determine the TCA enzyme activities affected by the increase in oxidative stress in children diagnosed with CP and to investigate the effect of these activity changes on the formation mechanism of CP by comparing it with the control group. In our study, 20 patients aged 4-18 years, who applied to the Pediatric Neurology Department of Bezmialem Vakıf University Hospital due to motor dysfunction and movement disorder, met the inclusion/exclusion criteria, were diagnosed with CP after neurologic, orthopedic and functional examination, and applied to the same hospital, but there was no evidence in their routine examination. A total of 40 patient serums, including 20 healthy children at the same age, whose disease has not been detected, will be included. In patients whose serum is taken; total oxidative capacity, total antioxidant capacity and Krebs cycle enzyme levels (alpha-ketoglutarate dehydrogenase, succinate dehydrogenase, fumarase, isocitrate dehydrogenase, glutamate dehydrogenase, malate dehydrogenase, citrate synthase) were measured. In statistical evaluations, t-test was applied to compare continuous variables between groups. Compared to the control group, alpha-ketoglutarate dehydrogenase enzyme level was found to be low in the patient group, the level of malate dehydrogenase enzyme was found to be high. (p<0.05). There was no significant difference between the groups in terms of other Krebs cycle enzyme levels, total oxidative capacity and total antioxidant capacity (P>0.05).
