Detection of Brucella and Leptospira by Multiplex Polymerase Chain Reaction in Cattle

Abstract (EN)
In this study, blood samples of 200 cattle in Aydın region were collected. After the extraction of DNA, the spesific genes related Brucella and Leptospira species were detected by multiplex PCR method and identificated. As a result of this multiplex PCR study for 200 samples, Brucella sp. and Leptospira sp. was detected from 77 (38.5%) samples. mPCR pozitive out of 77 samples, 24 (% 31.2)of them have shown bands both related to Brucella s.p and Leptospira sp. From the remaining 53 (% 68.8) samples, 33(% 62.3) of them have shown positive bands only related to Brucella sp. and 20 (% 37.7) of them have shown positive bands only related to Leptospira sp.As a result, the danger for contamination of samples and false statements in detection of Brucella and Leptospira by convantional methods and serum analysis methods were eliminated by multiplex PCR. In addition, trustworthy results would be obtained for identification of these two bacteria species without time expansions.
