Abstract (EN)
Aim : Shiga toxin (Stx), which is secreted by bacteria such as Shiga toxin producing Escherichia coli, Shigella diseanteria type 1 and Citrobacter freundi, increases the Interleukin (IL 1,IL 6, IL8 ) and Tumor Necrosis Factor (TNFα ) levels by causing renal inflammations in living organisms.By triggering oxidative stress, shiga toxin also causes to increase the level of Malondialdehyde and Nitric Oxide which are antioxidant parameters and by reducing Superoxide dismutase and Glutathione) levels, it causes to reduce the antioxidant capacity of the organism.This situation, especially by affecting kidney endothelial cells, results in Hemolytic Uremic Syndrome which causes symptoms such as microangiopathic hemolytic anemia, thrombocytopenia and acute renal failure.Strong antioxidant and anti-inflammation properties of the routine active ingredient are known. In the light of this information the aim of the study is to investigate the protective effects of routine active ingredients by using interleukins, tumor necrosis factor and axidative stress parameters on inflammation and oxidative stress in rats with Haemolitic Uremic Syndrome based on experimentally generated Shiga Toxin. Material and Method: In total, the study was conducted on 40 grown up male Sprague-Dawley rats weighing 160-250 gr at 12 weeks of age. Studies were performed in rats on HUS model generated with Shiga toxin 2 (Stx2) (100 ng / kg). By using two different doses(50mg/kg,100 mg/kg) and Interlocking agents, TNFα, oxidative stress parameters, biochemical renal parameters, histopathological methods protective effect of routine active ingredients on rats with Hemolytic Uremic Syndrome was evaluated. Results: After Stx2 (100 ng, / kg), Stx2 (100 ng / kg) + Routine (50 mg / kg), Stx2 (100 ng / kg) + Routine 100(mg/kg), Routine 100(mg/kg) doses were conducted, the levels of IL-1, IL-6, IL-8, TNFα, MDA, SOD, GSH, NO were calculated. Differences based on parameters between the groups were evaluated by one way ANOVA test. Conclusion: It has been concluded that Stx2 caused inflammation and oxidative stress in the kidneys and that the two different routine doses eradicated the effects of inflammation and oxidative stress.
