Diabetes mellitus (DM) is defined as chronic hyperglycemia due to insulin insufficiency, functional insulin disorders or both which causes disorders of carbohydrate, lipid and protein metabolism. DM is a disease which is associated with chronic complications. Diabetic noropatia is a chronic complication of DM that cause serious problems such as foot amputations, which negatively affects the quality of life in patients.In this study, we investigated the effect of spinorphin on painful diabetic neuropathy in streptozosin (STZ) induced diabetic mice.75 three weeks old male mice were used in this study. Following induction of diabetes in mice induced by STZ (nearly 4 weeks); Group one: Healthy control group (n=15). Group two: Diabetic control group (the group that spinorphin solver will be administered) (n=15). Group three: group DM+SP0.1 (the group that spinorphin solver will be administered 0.1 mg/kg). Group four: group DM+SP1 (the group that spinorphin solver will be administered 1mg/kg) (n=15). Group five: group DM+SP5 (the group that spinorphin solver will be administered 5 mg/kg) (n=15).Hyperglisemia levels which causes diabetic neuropathy, had been obtained in groups that STZ administered. Enough time for neuropathy progression had passed.When the Diabetic control group compared with Healthy control group, statistically significant difference was found in tersm of pain threshold (P<0.05). This result is admision for development neuropathy significant.Pain threshold values in DM+SP0.1 and DM+SP1 groups were not statistically significantly different in comparison to daibetic control groups (P>0.05).In DM+SP5 group, in 30 th and 45 th minutes pain threshold values for antinociceptive effect were statistically significantly different (P<0.05).In the study, we determined that spinorphine with dosage ?5mg/kg and intraperitoneal route in diabetic mice is effective in the acute antinociceptive treatment of painfull neuropathy.Key words: Diabetic neuropathy, Hot plate, spinorphine
