Abstract (EN)
ABSTRACTAimTo investigate the effect of granulocyte colony stimulating factor (G-CSF) given before 70% hepatectomy on liver regeneration in rats with steatotic liverMaterial-MethodThis study was performed in Laboratuary of Experimental Animals in Dokuz Eylül University School of Medicine. In the study, male Wistar-Albino rats at the average weight of 200-250 gr were used. The liver fattening model described by Lieber and Carli was used to generate liver fattening in rats.As a preliminary study, 3 rats were fed 70% fat rich diet during 8 weeks and then liver biopsy was performed to prove that the rats had sufficient level steatohepatitis. After we detected pathologically that the grades of steatohepatitis were 2 or higher in all 3 rats, we decided that the steatohepatitis model was performed succesfully.Three groups were chosen in the study:Group 1: G-CSF (Neupogen®, Roche, Amgen, Zug, Switzerland) at the dose 50 mcg (5MU) mixed with 5% dextrose was injected subcutaneously for 5 days before the 70% hepatotectomy to 7 rats which had grade 2 or higher steatohepatitis.Group 2: 5% dextrose solution at the dose 1 dzm was injected subcutaneously for 5 days before the 70% hepatotectomy to 7 rats which had grade 2 or higher steatohepatitis.Group 3: 5% dextrose solution at the dose 1 dzm was injected subcutaneously for 5 days to 7 rats which had no steatohepatitis.For biochemical evaluation, blood was taken from the tail vein just before the 70% hepatotectomy in group 1 and 2 and on fifth day of injection of 5% dextrose solution in group 3.In group 1 and 2, total hepatectomy was performed on the fifth day after the 70% hepatectomy and liver tissue was sent for pathological investigation of liver regeneration. Intracardiac blood was taken for biochemical investigation. Ki-67 proliferation index was evaluated pathologically to show liver regeneration.SPSS (Statistical Package for Social Sciences) for Windows 15.0 programe was used for statistical analysis.ResultsIn group 1, liver regeneration was statistically higher (p= 0,002). In the group which did not receive G-CSF, the levels of AST, ALP and direct bilirubine were statistically higher on postoperative fifth day. In Group 1, the levels of AST, ALT, ALP and total bilirubine were statistically higher. The increase of macro and microvesicular steatosis reduces the Ki-67 proliferation index.ConclusionG-CSF stimulates liver regeneration in rats with steatotic liver. The steatosis supresses the liver regeneration.
