Effects of grape seed extract on neuropathic pain in the streptozotocin induced-diabetic mice

Abstract (EN)

ABSTRACT Diabetes mellitus, a major health problem, is a syndrome that occurs with the microvascular and macrovascular complications triggered by hyperglycemia and chronic hyperglycemia in relation to the complete or partial failure of insulin secretion or insulin impact. Macrovascular complications, which are associated with atherosclerosis development, pave the way for the development of cardiovascular disease. Diabetic peripheral neuropathy is one of the major problems that reduces the quality of life and that is associated with morbidity and mortality. It affects more than 50% of diabetic patients and is characterized by loss of sensation, tingling and pain. Flavonoids, which are identified to have many positive effects on human health, have the characteristics of being antioxidant, anti-inflammatory, cholesterol lowering, anti-bacterial, antihyperglycemic, and anti-allergic. Being permeability regulator in the capillary microcirculation system and considering their effect of lowering blood pressure, they are called P factor or vitamin P as well. The main flavonoids are found in grape seed called proanthocyanidins. In this study, eight week old BALB / C strain mice were used. A total of 50 mice were used in this experiment consisted of five groups.To induce diabetes in thirty of these animals, single dose 180 mg / kg was administered intraperitoneally to be streptozotocin. After diabetes occurs, these groups (excluding healthy control group and diabetic control group), grape seed extract for six weeks 25mg / kg and 50mg / kg body weight was administered by oral gavage. At the end of the second and sixth weeks, a thermal pain model in these animals Hot Plate test by applying pain threshold measurements were performed. At the end of this study examined the tissue of the sciatic nerve and abdominal aorta from animals histologically. Grape seed extract-treated group and the control group were evaluated by comparing the results. Hot plate measurements in the group treated with grape seed extract and measurements between the other groups were considered significant (p<0.05). Grape seed extract administered in the groups, although diabetes-induced micro and macrovascular complications to be seen to reduce the damage assessment results were not statistically significant.
