Abstract (EN)
Tendons have their own fibroblasts (tenocytes) and these tenocytes form a dense extracellular matrix. Although it is the strongest tendon in the human body, the Achilles tendon is the most frequently injured tendon in the body. Many traditional treatments are used for tendon healing, including drug therapy, surgical treatment, and physical therapy applications. However, the desired results cannot be obtained with these treatment methods. Apart from these treatments, different treatment methods have been found with the development of regenerative medicine. These are two stromal vascular fractions (SVF) and bone marrow concentrate (BMC).The aim of this thesis is to evaluate the effect of using SVF and KIK on the treatment of Achilles tendon injuries. In the animal experiment created in the study, five animals were used for the control group and each experiment group. In this study, tendon healing was classified with a histological grading system. The tissue color and numerical data obtained by elastosonography were used in the biomechanical characterization of the healing tissue.As a result of the study stress ratios of 12 rabbits that received elastosonography were healthy group 0.30, control group 1.77, 100% SVF group 0.51, 100% BMC group 0.50, 50% SVF- 50% BMC group 0.69, 75% SVF- 25% BMC group was 0.62 and 25% SVF- 75% BMC group was 0.79 in first month. In the second month, healthy group is 0.23, the control group is 1.30, 100% SVF group is 0.35, 100% BMC group is 0.38, 50% SVF-50% BMC group is 0.33, 75%SVF-25% BMC group is 0.29 and 25% is SVF-% 75 BMC group is found to be 0.32. Histological evaluation was performed on 30 rabbit achilles tendons and mean scores were healthy group is 0, control group is 6.6, 100% SVF group is 9.33, 100% BMC group is 4.5, 50% SVF- 50% group is 5.5, 75% SVF-25% BMC group is 5.5 and 75% BMC -25% SVF group was found to be 7. With these results, it has been proven that the use of mixtures together improves the Achilles tendon more effectively than the individual uses.
