Abstract (EN)

Water is vital for the continuity of life. It is clear that water is necessary for the life system on earth and is an indispensable food source for human survival. Disinfection of water is carried out in order to consume safe quality water. Some disinfection by-products (DBP) are formed as an undesirable result of using chemical disinfectants in water disinfection. It has been determined in the literature that there are more than 700 disinfection by-products and pose a serious risk to human health. Some of the disinfection by-products have been reported to be cytotoxic, genotoxic, carcinogenic and teratogenic. In the thesis study, the cytotoxic effects of bromochloroacetonitrile in Haloacetonitrile (HAN) group, which is one of the various disinfection by-products formed after water disinfection, on the skin, which is the first barrier of our body in constant contact with water, bathing and swimming; MTT in HaCaT cells, the human keratinocyte cell line; It was investigated using LDH and ROS test methods. In addition, the effect of BCAN on wound healing in cell culture was investigated by the scratch test. As a result of the MTT test, it was determined that the 40 µM concentration of BCAN caused a decrease in the cell number of approximately 50% compared to the control after 48 hours of application. There was an increasing of LDH in the same time with the increasing of BCAN concentration. Increasing concentrations of BCAN have been observed to induce ROS production in HaCaT cells. It was determined that concentrations of 5,10 and 20 µM of BCAN did not have a negative effect on wound healing, but when toxic concentrations of 40, 60 and 80 µM were increased, it had a slowing effect on wound healing.
