Abstract (EN)

The frequency of cancer falls in the World and it occurs in phenotypic and genotypic conditions that occur in healthy cells. Because of uncontrolled cell proliferation, cells gain abnormalities and gain invasion and metastasis. Kidneys have factors related to metabolism and homeostasis is sufficient. Kidney cancer; urogenital cancers are involved and the incidence is increasing. In recent years, interest in plant-derived foods has increased and is often preferred to support traditional treatments. Throughout history, plant foods have been used in the treatment and bioactive components are also claimed to have pharmacological and physiological effects in the prevention of cancer. Chemopreventive compounds in dietary foods; by modulating the intracellular signal transduction pathways or prolonging the cancer formation process. Broccoli, cauliflower, white and black cabbage, Brussels sprouts such as Brassicaceae vegetables contain in the compound called glucosinolate. Glucosinolates react by catalyzing the enzyme myosinase. The disintegration product is a characteristic compound of isothiocyanate, sulforaphane (SFN). Due to its chemopreventive effects, SFN has received great interest in recent years. In vivo and in vitro studies with numerous epidemiological investigations have identified the potential of SFN to prevent or reduce cancer. The proapoptotic effects of SFN on various cell lines have been described, but there is insufficient study in the literature on the effect of apoptosis on kidney cancer. In this study, the effect of SFN on 769-P human kidney cancer cell line was investigated and 293-T human renal epithelial cell line was used as control group. Various SFN concentrations were prepared for this purpose and the effects on cell lines were examined.MTT [3- (4,5- dimethylthiazol-2-yl) -2,5-diphenyltetrazolium bromide] test (24, 48, and 72 hours) and LDH (Lactate dehydrogenease) for the detection of cytotoxic effect on cell proliferation test was applied. Cell cycle inhibitions were determined at the selected concentrations. In order to determine the apoptotic effect of SFN in cells, early and late apoptotic cells were analyzed by Annexin V method. In the 769-P human kidney cancer cell line, inhibition of cell migration in a dose-dependent manner was investigated.
