Isolation of active compounds from water extract of tanacetum alyssifolium plant and investigation of biological activities

Abstract (EN)

The aim of this study was to isolate the secondary metabolites present in the water extract of Tanacetum alyssifolium which is a member of Asteraceae family by chromatographic method (column chromatography), to determine the structure of the isolated compounds by spectroscopic methods, to investigate the anticarcinogenic effect and antioxidant activity. For this purpose, the Tanacetum alyssifolium plant was collected and dried under room conditions to avoid daylight. The plant was prepared with three different solvents: n-butanol, ethyl acetate, and water phase and the column was subjected to column chromatography. Two compounds were isolated from the water extract and their structure was determined by spectroscopic methods (1D-NMR and 2D-NMR). According to 1D-NMR and 2D-NMR results of the isolated compounds, it was determined that their structures were Routine and Isofracsetine. Antioxidant activities were determined using DPPH•, ABTS•+ free radical scavenging and chelating metal ions. Trolox and EDTA were used as standard antioxidants. Its antiproliferative activities are in ten different doses with Carboplatin, which is used as an anti-cancer drug on MCF-7 and MDA-MB-453 cell lines and examined. As a result, both compounds showed antioxidant activity, less than the standard structures, and antiproliferative activity was detected only in the compound of Isofracsetin, activity values were lower than the standard.
