Determination of some antibiotic residues in chicken meat by HPLC-DAD Method

In this study, side-by-side extraction of six antibiotics belonging to highly toxic four different classes, used in veterinary studies and frequently in chicken feed and water to ensure rapid growth of chickens, and determination of residue amounts by HPLC-DAD method and comparison of these residue amounts with maximum residue limit values is aimed. Chicken meat is one of the frequently consumed foods because of it is rich in essential amino acids compared to other meat products and affordable price. The residues of antibiotics are found in the tissues due to the excessive use for rapid production, incorrect use of drugs, and slaughter of animals without wating the time of antibiotics disposal from animals. It causes teratogenic, mutagenic, carcinogenic effects and increased antibiotic resistance in humans. The linearity of the developed HPLC-DAD method in the range of 0.05-100 mg L-1 is quite high in considering the regression coefficent (R2 = 0.9919-0.9999). The LOD values of the method for antibiotics other than penicillin G are in the range of 6-27 µg L-1 and LOQ values are in the range of 20-89 µg L-1. For penicillin G, the LOD value is 1.56 mg L-1 and the LOQ value is 5.21 mg L-1. The accuracy is between 26,50 and 13,20% relative error. The precision in terms of retention times, measured concentration and peak areas is in the range of 0.01-18.10 % relative standard deviation. Recovery values for the extraction procedure ranged from 73.51-113.17 % for the six antibiotics. These results indicating that the HPLC-DAD method is reproducible for determinations. As a result of HPLC-DAD analysis of chicken breast meat samples of 11 different companies, chloramphenicol residue lower than the LOQ value was detected in 1 sample. Chloramphenicol is one of the banned antibiotics and has no maximum residue limit value. In 1 sample, oxytetracycline lower than the LOQ value was detected. The result obtained is below the maximum residue limit values and is within the legal limits. HPLC-DAD results were confirmed by LC-MS/MS and there was no significant difference between the results according to the t test result.
