EFFECTS OF ACETYLCYSTEİNE ON APOPTOSİS AND TRPM2 CATİON CHANNELS EXPRESSİON İN TORSİON-DETORSİON CREATED RAT TESTİS TİSSUE Testis tortion is a rarely seen urgent urologic pathology, which gives rise to serious irremediable fertility problems if it is not treated on time. In this work, the investigations made on the effect of a well-known antioxidant N-Acetylcysteine (NAC) compound on prevention of the ischemic destructions, which arises from testis tortion-detortion steps. İn the study,35 (Spraque-Dawley) male rats were preparation chosen with the same species and ages (8-10 weeks old). Then all the rats divided into 5 groups with the same number. Group I: Control group (n:7); in the experiments, not any process was performed on those 7 male rats in order to make comparison with the other groups. Group II: Sham group (n:7); the abdomen was surgically cut, and the left sides of the testis were taken out and then keep them waiting for 60 min. without performing anymore process. After that, the left sides of the testis were replaced into the abdomen and keep them initial positions. Group III: NAC group (n:7); single dose (100 mg/kg) of NAC was applied as intraperitoneal (i.p.) to the 7 male rats. Group IV: Tortion-detortion group (n:7); the abdomen was surgically cut, and the left sides of the testis were taken out, then the tortion was performed by rotating it 7200 towards the counter clock wise. The tortion has completed in 60 min., after this time, detortion was performed and the left side of the testis was replaced to the abdomen and keep it initial position. Group V: Tortion-Detortion + NAC group (n:7); the abdomen was surgically cut, and the left sides of the testis were taken out, then the tortion was performed by rotating it 7200 towards the counter clock wise. The tortion has completed in 60 min., after this time, single dose (100 mg/kg) of NAC was applied as intraperitoneal (i.p.) to the 7 male rats. After that detortion was performed and the left sides of the testis were replaced to the abdomen and keep them initial positions. After 48 hours, the surgical work has completed and the rats in all the groups were killed (headless method) under the anaesthesia. Routine histological tissue preparation methods.Tissue sections were stined by using HE,PAS and Massons frichnome techniques. Moreover, tissue sections were stained by using Audun Brotin Peroksidaze technique and TUNEL methods to determine TRPM2 immünereativity and apoptosis respectively. The results of Group II – V (tortion-detortion applied groups) were compared with the control group (Group I) by using an optic microscope. According to the comparison, the following results obtained in tortion-detortion groups, • degeneration was determined in seminiferous tubule germinal epithelium, • separation was monitored in seminiferous tubule basal membrane, • oedema was determined in atrophic tubules and interstitial area, • and also, it was observed that immature cells decanted in some lumens of tubules. As to tortion-detortion + NAC applied group, an apparent recovery was observed in testis tissue, degenerated germinal epithelium, vascular congestion, and interstitial oedema. Also, a decrease was determined in the separation of seminiferous tubule basal membranes. When comparing the tortion-detortion group with the control group, a meaningful increase was determined in TUNEL positive cells. As to tortion-detortion + NAC groups, it was determined that the number of TUNEL positive cells was almost similar with the control group. In terms of TRPM2 immunoreactivity, the tortion-detortion group was compared with the control group and the results showed a meaningful increase. However, in trortion-detortion+NAC group, a decrease was observed in terms of TRPM2 immunoreactivity. According to the control group, the value of TOC was statistically getting increase, but, the value of TAC was getting decrease. So it can be say that the application of NAC put the value of TAC and TOC in a close aproximate with the value of control group. As a conclusion, it can be said that the application of tortion-detortion has an detrimental effect on the male reproductive system. NAC was chosen for the purpose of preserver against the tortion-detortion due to having antioxidant feature. The results supported our purpose with showing a significant decrease in unfavourable effects of tortion-detortion. Key words: Tortion, N-Acetylcysteine, Rat, Testis, Tunel, İmmünohistokimyasal
