The relationship between tuberculosis susceptibility iIL-17A, IL-17f and IL-23R gene polymorphisms and İrak-1 and İrak-3 innate immune regulators

The susceptibility to tuberculosis (TB) and prognosis of the disease depend on the interactions between the bacterial agent, the host immune system, environmental and genetic factors. Although studies have shown that cytokine gene polymorphisms are associated with TB, their mechanisms of action have not been fully elucidated. In this case-controlled study, we aimed to investigate the role of single nucleotide polymorphisms in IL-17A, IL-17F and IL-23R genes in susceptibility to TB and the relationship of human innate immune response to TB susceptibility. The TB group was comprised of 50 individuals, 31 (62%) male and 19 (38%) female in the study. The healthy control group was comprised of 50 individuals, 21 (42%) male and 29 (58%) were female. When the gender distributions of the patient and healthy control groups were compared, a significant difference was found (p=0.045). The mean age of the healthy control group participating in the study was 37.34±11.519 and the mean age of the patient group was 45.82±13.002. When the mean age of the patients and controls was compared, a statistically significant difference was found (p=0.001). When smoking habits were evaluated between the patient and control groups, a high rate of 70% was found and a statistically significant difference was found when compared (p=0.001). Blood samples were taken from the patient and control groups in EDTA tubes. DNA extraction was performed first from the blood samples. Then, polymorphisms in the IL-17A gene (rs2275913) and IL-17F gene (rs763780) were investigated by using Amplification Refractory Mutation System-Polymerase Chain Reaction (ARMS-PCR) assay. The presence of polymorphisms in the IL-23R gene (rs11209026) was investigated by using Polymerase Chain Reaction-Restriction Fragment Length Polymorphism (PCR-RFLP) assay. After mononuclear cell isolation from the remaining blood samples, RNA extraction and cDNA extraction from the obtained RNAs were performed. Then, the mRNA levels of IRAK-1 and IRAK-3 innate immune response regulators, which are among the innate immune response regulators, were analyzed by quantitative Real-Time PCR method. After obtaining all the results, the relationship between gene polymorphisms and IRAK-1 and IRAK-3 regulator genes was statistically compared. There have been many studies investigating the relationship of IL17/IL23 with different diseases. However, our study is the first study in our country to investigate the relationship with tuberculosis. We believe that the data of our study will be very important in terms of literature for different results found in various countries and will be a reference for new diagnosis and treatment studies.
