Evaluating reliability of SE33 location in forensic DNA identification studies in turkish society

Abstract (EN)

Aim: In our country in forensic STR (Short Tandem Repeat) DNA loci are increasingly used more day by day in recent years. Genotype similarity rate in Turkish population is higher than in western populations. Therefore, more and more reliable autosomal STR loci are needed in identification studies. In this context, it is aimed to determine the polymorphism rates, frequency, matching frequency and to obtain information of SE33 region. Material and Methods: This is a retrospective study. Between January and June 2018, the investigation was carried out by the Judges and Prosecutors' Offices to give samples to İzmir Forensic Medicine Group Presidency Biology Specialization Department or by taking blood, hair, saliva etc. samples taken from public hospitals. In this context, evaluations were made on samples belong to 500 individuals who did not have close relatives. DNA isolation kits (EZ-1 investigator kit and Power Plex Fusion 6C Direct kit) were used to isolate genomic DNAs. The isolated DNAs are then amplified by the SE33 STR locus in combination with the Power Plex Fusion 6C set. Reasulst: In the study, homozygous genotype ratio was calculated as 5.2%, heterozygous genotype ratio was 94.8%, discrimination power (PD) was 0.992 and probability of match (PM) was calculated as 0.008. The lowest frequency alleles were determined as 0.1, 6, 7, 10, 16.3, 17.2, 23, 24, 29, 34.2, 35.2, 37 alleles. Conclusions: In the light of the above information; The SE33 STR locus indicates that the discriminating power and probability of matching may be a strong marker in forensic identification when compared to other studies.
