Phylogenetic analysis of the most commonly cultivated local and foreign cotton varieties in Turkey using SRR markers

Abstract (EN)

Objective: This study was conducted to reveal the genetic relationships of cotton varieties cultivated in Turkey. Material and Method: The study was conducted by obtaining a total of 30 cotton varieties from Nazilli Cotton Research Institute Genetic Stock and registered institutions. Samples were taken from the seeds by crushing and grinding. DNA isolation was performed in the laboratory according to the kit protocol and genetic relationships were revealed with 10 SSR markers. Findings: The data obtained in the first part of the study were realized on cotton DNA quantity and purity. In the study, amplification was performed using 10 SSR primers for 30 cotton genotypes and a total of 39 bands were obtained. 8 of them were monomorphic and 31 were polymorphic. The polymorphism rate was determined to be approximately 79.48%. DNA agarose gel images were scored and a Phylogenetic tree was revealed in the Past4.06 Project program. Conclusion: In this study, it was determined that there is a large gene pool for future cotton breeding studies due to the high polymorphism of genetic diversity.
