Wound healing complicated process including different cells, hormones and growth factors. In last decades, There has been a lot of commercial products about the wound dressing including, alginate, collagen, chitosan etc. In this study, MADOPA-containing hydrogel was developed to contribute collagen-based wound healing. Collagen was isolated from the sheep skin by pepsin extraction method and charecterized with Sirius red quantification, Uv-vis spectroscopy, CD spectroscopy, SDS-page and FT-IR-ATR. Tyrosinase enzyme from edible mushroom (agaricus bisporus) was partialy purified by exraction, ammonium sulfate precipitation and sephadex g-75 gel filtration for the production of MADOPA, which is the other composition of hydrogel. Charecterization of tyrosinase was performed with FT-IR, SDS-page, BCA quantification and tyrosinase activity determination. Third part of the working is devolepment of immobilise tyrosinase system with poli (HEMA-MAH-Cu (II) cryogel column. Swelling test, FT-IR, SEM monitoring and yield of the column was performed for charecterization. Transformation of MAT to MADOPA is the fourth part of the working. In this step, diffarences between free and immobilise enzyme was compared with the production of MADOPA. Final part of working was production of collajen-co-DOPA-co-MATrp-Ru(bipyr)2-Cl with 3D hidrogel for wound dressing. This material is biodegradable, biocompatible and non-toxic.
