Urinary excretion of 8-hydroxy-2’-deoxyguanosine in patients undergoing surgery for urological cancer

8-hydroxy-2?-deoxyguanosine (8-OHdG) is the most frequently detected and studied DNA lesion. Upon DNA repair, 8-OHdG is excreted in the urine. Urinary 8-OHdG is now considered as a biomarker of generalized, cellular oxidative stress and is linked to degenerative diseases including cancer. In this study it is aimed to analyze efficiency of assaying urinary excretion of 8-hydroxy-2?-deoxyguanosine in patients undergoing surgery for urological cancer (bladder, prostate and renal cell cancer (RCC)). We have also assay groups including benign prostatic hyperplasia (BPH), healthy control and the patients with renal stone. Urinary samples were studied with highperformance liquid chromatography and electrochemical detector (HPLC-EC). Group 1 included cancer patients (bladder, prostate and RCC) with an average of 8-OHdG/creatinine rate (0,143 ± 0,097) is sequentially found to be higher than control, BPH and renal stone groups (0,084 ± 0,036, 0,067 ± 0,017, 0,067 ± 0,029, p<0,05). On the other hand, no significant difference is observed within control group, BPH and renal stone patients. Furthermore, bladder and prostate cancer patients? 8-OHdG/creatinine rates has found to be higher than control group (p<0,05). Within 14 patients undergoing surgery 12 (85,7%) had decreasing values of 8-OHdG/creatinine rates, indicating a statistical significance (0,135 ± 0,093 & 0,092 ± 0,034, p<0,05). As a result of our findings, it is concluded that 8-OHdG/creatinine rates are potential biomarkers for detecting oxidative DNA damage in cancer patients. 8-OHdG/creatinine rates can also be used as an indicator of surgical therapy.
