Contribution of WR 2721 (Amifostin) to the Antioxidant and Hepatoprotective effects of UW (University Of Wisconsin) and HTK (Histidine-Tryptophan-Ketoglutarate) preservation solutions

Abstract (EN)

In liver transplantation, as in all organ transplantation programs, ito reduce ischemiareperfusion injury during organ preservation, solutions such as Histidine-Tryptophan-Ketoglutarate.(HTK).and.University.of.Wisconsin.(UW).were.introduced. Since 1969, amifostin has been used in order to protect the cells from the damaging effect of radiotherapy and chemotherapy. Amifostin`s cytoprotective effect has been proved in many clinical trials. In our study we aimed to analyze the additive effect of amifostin.to.the.cytoprotective.effect.of.HTK.and.UW. In this study the 48 rats were subclassified into 6 groups. For preservation, we used Ringer's Lactate (RL) in the first group, RL + amifostine in the second group, HTK in the third group, HTK and amifostine in the fourth group, UW in the fifth group and UW + amifostine in the sixth group. Two hundred mg/kg of amifostin was applied subcutaneously 30 minutes before hepatectomy. After perfusion, hepatectomy was applied and the tissue preserved in the same +4oC. At the 0'th, 6'th and the 12'th hours, tissue was obtained from liver and fluid at perfusion fluid respectively. Biochemically Alanine aminotransferase (ALT), alkaline phosphatase (ALP) and nitric oxide (NO), immunohistochemically Inductible Nitric Oxide Synthase (iNOS) and via terminal dUTP nick end labeling (TUNEL) method apoptosis were analyzed. Additionally, pathologic changes which have been occurred in the cells were analyzed via light.and.electron.microscope. As a result, in the groups which amifostine was used hepatocyte damage was dramatically reduced after the sixth hour, especially in the UW perfused group. In the light of electron microscope analyses, the cells architecture was best preserved in UW + amifostine group. The architecture was more impaired in groups which amifostin has not been.used. In the light of these findings, we can say that amifostine is the potent agent when it's used before hepatectomy with preservation solutions.
