Abstract (EN)

Methicillin-resistant Staphylococcus aureus (MRSA), which has increased in our country in recent years, is frequently confronted with nosocomial infections. Vancomycin and daptomycin are of great importance in the treatment of antibiotics used for MRSA with a high mortality rate. However, after the discovery of vancomycin in the 1950s, over time, there has been a tendency to find alternative antibiotics for many reasons such as resistance to vancomycin, application frequency, intense pain during application and, rarely, red man syndrome. The use of daptomycin, marketed in conjunction with the approval of the FDA in 2009, has quickly gained popularity due to the single dose daily, painlessness and low resistance development. However, it was observed that both antibiotics showed nephrotoxicity in a certain proportion of patients. The aim of this thesis study is to determine the ratios of renal damage caused by administration of vancomycin and daptomycin to rats for 2 weeks using related parameters. 21 male Sprague Dawley rats weighing 200-250 g, 8 weeks old were used. The rats were divided into 3 groups with 7 animals in each group. 1. group (control group) was administered with saline, group 2 vancomycin 200 mg / kg, group 3 daptomycin 4 mg / kg once daily intraperitoneally for 14 days. Twenty-four hours after the end of the injections, the rats were sacrificed. CR (creatinine) and urea from urine specimens obtained from rats, BUN (blood urea nitrogen) and CR values were determined from blood samples taken from rats. Creatinine clearances were calculated from these values. NAG (N-acetylglucosaminidase), CAT (catalase) and GSH (reduced glutathione) enzyme activities and MDA (malondialdehyde) levels were detected in kidney tissues. In addition, renal tissue was examined histopathologically and immunohistochemistry revealed apoptotic cells and findings in the kidney. At the end of our study, kidney damage was observed in rats administered vancomycin and daptomycin. In the control group, kidney damage was not observed. However, we found that vancomycin had a higher level of renal damage than daptomycin.
