The effects of vermicompost application on phenypropanoid metabolism of basil (Ocimum basilicum L.) chemotypes

Abstract (EN)
In this thesis, the effects of solid vermicompost (KV) and liquid vermicompost (SV) applications on phenylpropene accumulation and expression profiles of PAL, 4CL, EGS, EOMT and CVOMT genes in peltate glandular trichomes isolated from young leaves of methylcavicol chemotype (LR10A) and methyleugenol chemotype (LR25A) basils were investigated. For this purpose, 0%, 10% and 25% KV to root regions of chemotypes; pure water, 10% and 25% concentrations of SV were applied to the leaves by spraying. It was determined that KV applications decreased the accumulation of methylchavicol and methyleugenol and the expressions of EOMT and CVOMT genes in methylchavicol chemotype basils, whereas increased them in methyleugenol chemotype basils (p<0.05). The application of 10% SV to methylchavicol chemotypes increased the percentage of chavicol and methyleugenol, and 25% SV application increased the percentage of eugenol and methylchavicol. The application of 10% SV increased PAL, EOMT and CVOMT gene expressions, whereas 25% SV application generally decreased gene expressions. SV applications to methyleugenol chemotypes decreased the ratio of eugenol and methyleugenol, but did not cause a significant change in the ratio of methylchavicol (p<0.05). In methylogenol chemotype basils, SV treatments increased the PAL gene expression in direct proportion with the increase in dose, while it had different effects on the expressions of other genes. As a result, significant differences were found in phenylpropene content and gene expression profiles in the peltate glandular hairs, depending on the form and dose of the applied vermicompost and the chemotypic differences of basils.
