Pectin Methylesterase Inhibitor (PMEI) genes have a very important place for plants. They are involved in many different biological processes in plants such as stress response, plant development, fruit formation and maturity. In this study, PMEI genes in grapevine (Vitis) were examined in silico through bioinformatic analyses. 46 genes in Vitis vinifera and 43 genes in Vitis riparia were identified as PMEI genes and were named as VvPMEI and VrPMEI. It was observed that the motif patterns of the proteins in the same class were similar in the phylogenetic trees found. VvPMEI and VrPMEI targeting miRNAs were found and the relationships of these genes with stress response were tried to be determined. VvPMEI-10, VvPMEI-18, VvPMEI-34 and VvPMEI-35 genes were found to be potential genes related to salt, cold and drought stress, and potential target genes related to fruit formation, coloration and ripening were determined. In addition, the FastPCR program was used to investigate in-silico whether the changes in gene expression levels in grapevine gene sequences exposed to salt stress obtained from databases detected and it was seen that it could determine the changes in gene expression. As a result, it is thought that this study can be a source for further research and breeding studies. It was determined that in-silico studies have many advantages such as saving time and cost in the development of varieties resistant to salt stress and other abiotic stresses.
