Özet (EN)

In this study; 2-acrylamido-2-methyl-1-propane sulfonic acid (AMPS) based cationic exchanger cryogel column has been developed for the purification of horseradish peroxidase (HRP) by ion exchange chromatography, the efficiency of this column for HRP purification has been investigated. Then, purified HRP cryogel column has been developed and poly(3,4-ethylenedioxythiophene) (PEDOT) conductive polymer has been synthesized by enzymatic polimerization method by using this column. In this direction in the first stage; AMPS cryogel column has been prepared by the polymerization of hydroxyethyl methacrylate (HEMA) monomer in the presence of N,N' Methylenebisacrylamide (MBA). Characterization of the prepared cryogel column has been carried out by swelling tests, BET, SEM and FT-IR analysis, Bradford method for protein content of the purified HRP and SDS-Page analysis for determination of molecular weight. In the second step of the study, HRP cryogel column has been prepared by MBA, HEMA and ruthenium based amino acid monomers using photosensitive cross-linking technique (ANADOLUCA). Then, PEDOT/PSS polymer has been synthesized from HRP cryogel column by poly(sodium 4-styrenesulfonate) and 3,4-ethylenedioxythiophene monomer by using peristaltic pump. Molecular weight of synthesized polymer has been determined by Gel Permation Chromatography (GPC).
