Abstract (EN)

In this study, a spectrophotometric method has been developed for the determination of Cu+2 and Ni+2 ions in biological fluids by using 2,4 disubtitüe thiazole derivate Schiff bases, 4-(1-mezitil-1-metilsiklobütil-3-il)-2-(2-hidroksibenzilidenhidrazino) tiyazol (L1) and 4-(1-fenil-1-metilsiklobütil-3-il)-2-(2-hidroksibenzilidenhidrazino) tiyazol (L2) ligands. Optimum reaction conditions along with other analytical parameters of the developed method in the application on biological fluids were determined which were used in the analysis.In the first stage of the study, the coloured complexes of Copper (II) and Nickel (II) ions with L1, L2 ligands were formed in aqueous media. Since the resulting complexes are not soluble in water, it extracted into chloroform and the absorption of the solution were measured from this solutions in each case. The maximum absorption wavelengthies for the complexes were determined from their organic solutions. All the determinations were carried out at the determined wavelengthies throughout the work. The method was optimized for pH, amount of reagents required, time, mol ratios, interfering species and buffer for the complexes.In the second stage of the study, calibration curves complexes were obtained respectively. The molar absorptivity, specific molar absorptivity, Sandell?s sensitivity and relative standard deviation were determined.The developed method for Copper and Nickel determination in biological fluids was compared to Atomic absorption spectrophotometric method by using Standart Referans Material (Metals in human serum, Medisafe® Metalle S, Level 1, ME 28341). The developed method seems to be a sufficient route to determination of copper and nickel amounts in biological samples.Keywords: Schiff bases, Thiazole, Copper, Nickel, Complexes, Spectrophotometry.
