Abstract (EN)

In this thesis, a biosensor modified with reduced graphene oxide based bimetallic platinum-cobalt nanoparticles (Pt-Co@rGO NP) has been developed for the selective and sensitive detection of AA, DA and UA. AA, DA and UA are bioactive molecules that play key roles in the human body. Pt-Co@rGO nanoparticles were synthesized using a microwave assisted technique. Characterization of Pt-Co@rGO nanoparticle was examined by TEM / HR-TEM, XPS, XRD and Raman spectroscopy. Characterization results showed that this biosensor has excellent properties such as mechanical strength, large surface area, stability and superior conductivity. The well separated AA, DA and UA oxidation peaks of the Pt-Co@rGO biosensor have been demonstrated by DPV and CV measurements in the range of -0.2 V to +0.8 V. The prepared Pt-Co@rGO biosensor showed high electrochemical activity, a large linear range, high sensitivity and acceptable detection values for individual and simultaneous detection of AA, DA and UA biomolecules under optimized conditions. The linear range of Pt-Co@rGO for AA, DA and UA respectively 170-200; 35-1500 and 5-800 µM. Moreover, the limit of detection of the prepared biosensor for AA, DA and UA, respectively 0.345; 0.051; 0.172 µM. As a result of these studies, this non-enzymatic Pt-Co@rGO based biosensor has proven to be a promising, ideal biosensor for future detection of neurodegenerative diseases.
