Determinaion of the genotoxic profiles of new synthesis oxipropanolamin derivatives

Abstract (EN)

The aim of this study was to determine the genotoxic and cytotoxic profile of the 1-(2-isopropyl-5-methylphenoxy)-3-((pyridin-3-ylmethyl)amino)propan-2-ol, which is newly synthesized in Sakarya University Chemistry Department. Chemical substances which are considered as pharmaceutical raw materials are subjected to a wide variety of toxicological researches. Some of these studies are cytotoxicity and genotoxicity studies In this context, chromosomal abnormality (CA) and micronucleus (MN) tests were performed to determine the cytotoxic and genotoxic effects of test substance on in vitro human peripheral blood lymphocytes. For this purpose, the lymphocyte culture was treated with four different concentrations of test substance (6.25, 12.5, 25, 50 μg/mL) for 24 and 48 hours. According to the data obtained in the study, it was observed that the test substance significantly reduced the mitotic index in all application groups compared to the negative control. According to the solvent control, significant reduces were observed in the concentration of 25 and 50μg/mL in both 24 and 48 hours of application. The most observed chromosomal abnormality was chromatid fracture in all groups, followed by fragment and chromosome fractures. Significant increases in the maximum two concentrations were observed with respect to the percentage of abnormal cells and chromosomal abnormalities per cell. For both evaluation criteria, significant increases were observed in the highest concentration of solvent control in both application periods. However, in terms of chromosomal abnormalities per cell, a significant increase in the concentration of both 25 and 50 μg/mL was observed in 48 hours. In terms of micronucleus frequency, there is a significant increase in all concentrations and in 50 μg/mL wasn't seen any binucleats.According to the results obtained, the applied concentrations of the test substance were cytotoxic in in vitro conditions and genotoxic effect, especially at high concentrations.
