Abstract (EN)

Aim: In this thesis study, studies were carried out to determine the inhibition effects of the newly synthesized Schiff base ligand (LH) and metal complexes (CoL2, CuL2, NiL2 and ZnL2) as an alternative to Acetazolamide used in the treatment of Glaucoma, on the activities of hCA I and hCA II isoenzymes. Material and Method: In this thesis study, the inhibition effects of the synthesized Schiff base metal complexes on hCA I and hCA II isoenzymes were examined. Acetazolamide was used as a positive control. IC50 ve Kİ values were calculated for each metal complex and inhibition types were determined. Result: In the study, IC50 ve Kİ values were determined to be in the range of 111.70 ± 2.18-771.90 ± 17.55 nM and 99.16 ± 9.21-721.90 ± 53.46 nM, respectively, for the hCA I isoenzyme. The same parameters were calculated for the hCA II isoenzyme in the range of 160.50 ± 3.27-1076.00 ± 29.39 nM and 125.60 ± 9.31-1033.00 ± 56.99 nM, respectively. Conclusion: It has been determined that the CoL2 compound, one of the Schiff Base metal complexes we used in the study, significantly inhibits the hCA I enzyme.
