Abstract (EN)

Aim: Within the scope of this thesis, it is aimed to determine the kinetic parameters of the newly synthesized Schiff Base ligand (LH) and metal complexes (CoL2, CuL2, NiL2 and ZnL2) as an alternative to the drug tacrine used in the treatment of Alzheimer's disease, on the activities of AChE and BChE enzymes. Material and Method: AChE and BChE activities of the compounds were determined spectrophotometrically by the Ellman method. Inhibition types were determined by calculating the IC50 values and Ki constants of the newly synthesized Schiff Base ligand (LH) and metal complexes (CoL2, CuL2, NiL2 and ZnL2), which showed in vitro inhibition effects for AChE and BChE. Result: While the KI values of Schiff Base ligand (LH) and metal complexes (CoL2, CuL2, NiL2 and ZnL2) for AChE vary between 26.94 ± 2.14 and 65.85 ± 5.34, for BChE it is 345.10 ± 30. It was determined to be between 31 and 1187.00 ± 62.23. Conclusion: Schiff It has been determined that some metal complexes significantly inhibit AChE and BChE enzymes. 2024, 66 Pages Keywords: AChE, BChE, Schiff Base Metal Complexes.
