Abstract (EN)
OBJECTIVE: Ovarian cancer is the second most common gynecologic malignancy and the most common cause of gynecologic cancer death. High-grade serous ovarian cancer (HGSC), accounting for approximately 70% of epithelial ovarian canser, is an aggressive ovarian cancer that associated with a worse clinical outcome. It is a poor-prognosis tumor with a 5-year survival rate of around 40%, which is due to late diagnosis (75% in FIGO III/IV) as well as the development of resistance to chemotherapy. Ubiquitination is widely recognised as a fundamental regulator of numerous cellular events, indeed the role of ubiquitination in DNA damage response, cell cycle regulation and intracellular trafficking has been extensively characterised. Consequently, much attention has been focused on determining the association between ubiquitination and tumorigenesis. Ubiquitin Specific Protease 17 (USP17) and found that it regulated signalling through the Ras pathway by controlling the intracellular localisation of Ras and other small GTPases; key regulators of cell proliferation and migration. Tumor protein translationally controlled 1(Tpt1), also known as translationally controlled tumor protein (TCTP), is an evolutionary conserved protein that shares a high degree of homology from plants to mammals, implicating its fundamental functions. Tpt1 is associated with cancer progression and is highly expressed in many tumors. Recently, Tpt1 was found to interact with p53 and to negatively regulate p53 by increasing its degradation. MATERIAL-METHOD: The study examined the results of staining with USP17 and TPT1 receptor kits on pathologic blocks of patients diagnosed with high-grade serous ovarian carcinoma due to who had operation because of adnexal mass between June 2013 and June 2017 and definitive pathology results. The rate of receptor positivity of these patients in primary tissue and metastatic tissue was investigated. It was planned to investigate the relationship between receptor positivity and metastasis. DISCUSSION: Previous studies of USP17 and Tpt1 expressions evaluated in different types of cancer, we evaluated in primary and metastatic tumor tissues of patients with high grade serous over cancer and increased expression was observed in both groups. Although the increase was considered to be associated with tumor progression and metastasis statistically significant difference was not obtained due to low number of patients. CONCLUSIONS: There was a statistically significant difference between primer and metastatic tissue expressions of USP 17 and USP 17-related immunohistochemical stainings and expression of primer over tissue and metastatic tissue. The correlation coefficient 0.349 p = 0.047, although there was a correlation it was weak, but Tpt 1 immunohistochemical staining showed no statistically significant correlation.
