Abstract (EN)
In this study, thermo-tolerance of Ganoderma lucidum monokaryon, obtained by spor germination, has been determined against high temperature and the comparison of strains has been accomplished by RFLP (Restriction Fragment Length Polymorphism) analysis of extracted DNA from Turkey isolates by using EcoRI enzyme. Ganoderma lucidum spores were spread-inoculated on solid CYM (Complete Yeast Medium) by diluting. Single germinated spores after incubation at 25ºC, subcultured to another CYM. These monokaryon has then been tested for determination of tolerance to temperatures ranging from 25ºC,32ºC, 35ºC, 37ºC to 40ºC. The highest growth rate was seen at 27ºC and 32ºC temperatures. Although there is no growth obtained at 40oC, fairly decreased growth rate were seen at 35oC and 37ºC compared to 27ºC and 32ºC. By applying chromasomal separation ,one of the most reliable genom analysis, and total genomic size determination technique on different Ganoderma sp strains, it was found that this Basidiomycetes contain chromosomes in a few mb size, only two chromosome could be 200 kb size .
