The animal models are often used to investigate new strategies against diabetes, which is a rapidly increasing ailment worldwide. High-fat diet (HFD) and streptozotocin (STZ) administration in rodents is a suitable model of human type 2 diabetes (DM2). The protective or therapeutic effects of various phytochemicals have been investigated against DM2 and its complications. Curcuma amada Roxb (Mango Ginger, MG) is well-known for its pharmacological and biological effects such as antioxidant and anti-inflammatory etc., in the field of medicinal plants. The aim of this study is; assessing the effects of MG extract which may have potent protective effects against the adverse effects of diabetes in high fat diet (HFD) and streptozotocin (STZ) induced diabetic rats, and it is exhibited how MG effects GSK-3 beta/Fyn /Nrf2 signaling pathways. A total of twenty-eight Wistar albino rats divided into four groups; i) Control group (non-diabetic rats) fed a regular diet; ii) MG group rats fed regular diet and administrated with MG (50 mg/kg BW per day); iii) HFD+STZ group rats fed an HFD and then injected STZ (40 mg/kg, i.p.); Group IV rats fed an HFD and administrated with MG (50 mg/kg BW); and iv) HFD+STZ+MG Group rats fed HFD and then injected STZ administrated with MG. The study conducted for 12 weeks. Blood glucose, insulin, and some biochemical values increased in diabetic rats (P<0.01), however, MG administration alleviated this condition (P<0.001). Serum and liver malondialdehyde (MDA) increased (P<0.001) and total antioxidant capacity (TAK) decreased in diabetic rats (P<0.001). In the YYD + STZ group, GSK-3β, Fyn, and Keap-1 protein levels increased while Nrf2 and HO-1 protein levels decreased, whereas MG administration had an adverse effect in diabetic rats (P<0.05). In diabetic rats, MG treatment improves biochemical parameters and regulates the GSK-3 beta / Fyn / Nrf2 signaling pathways, thereby improving the antioxidant defense mechanisms and thus alleviating supportive and complications in the treatment of diabetes.
