Development of new ISSR primers from pistachio SSR loci

Abstract (EN)

In this study, it is aimed to develop new ISSR primers from pistachio SSR loci using the genomic libraries enriched with four different repeats by testing them in pistachio, walnut, apple, apricot and in cherry. In the present study, eight varieties or genotypes from each of five species and 137 newly designed ISSR primers were used. Thirty-six ISSR primers from University of British Columbia (UBC) were also tested for the comparison. From new ISSR primers, a total of 791 fragments were amplified and 618 were polymorphic in pistachio, a total of 613 fragments were amplified and 395 were polymorphic in walnut, a total of 696 fragments were amplified and 496 were polymorphic in apple, a total of 666 fragments were amplified and 429 were polymorphic in apricot, a total of 514 fragments were amplified and 281 were polymorphic in cherry. Number of total bands and polymorphism levels of new ISSR primers were similar to the UBC primers. As a result, this study showed that new ISSR primers developed in this study can be used for cultivar identification, characterization of genetic resources, marker development and genetic mapping studies.
