Abstract (EN)

The promoter gene for tumour necrosis factor alpha (TNF-) can control plasma levels of TNF-, which are frequently observed in RA. A C850T (rs1799724) polymorphism in the promoter region of the TNF- gene was the primary goal of this study, which attempted to investigate the pathophysiology of RA. A total of 100 participants were included in the current study, who were split into two groups: 50 had been diagnosed with RA, while the other 50 appeared to be in good health. The participants' ages ranged from 18 to 70. (17-30 years). ELISA was also used to determine the level of TNF- in the body. Polymerase chain reaction (PCR) was used to amplify TNF- using particular published primers. The restriction enzyme Hind II was used to digest the PCR result. The present study found numerous significant differences between RA patients and healthy individuals. In comparison to controls, all patients exhibited higher BMIs (27.70± 5.07), waist-to-hip ratios (0.85± 0.04), and TNF- levels (23.19 ±3.95) than expected (P=<0.001). As a result of the increased body weight and subsequent insulin resistance, the current research suggests that TNF- may be involved in RA pathogenesis. While a polymorphism of the TNF- C850T (rs1799724) in the examined population did not influence the development of RA.
