Abstract (EN)
AİM:The purpose of the study is to investigate the antioxidant and anti-inflammatory effects of high-dose and short-term betaninadministration after colitis is established in an experimental ulcerative colitis model.With the results to be obtained, it is aimed to determine the effectiveness of acute administration of betanin in eliminating oxidative damage and inflammatory response caused by ulcerative colitis.If this application gives positive results, it is thought that betanin will acquire qualification for ulcerative colitis patients during attack periods. Material - Method: With the approval of Aydın Adnan Menderes University "Animal Experiments Ethics Committee", a total of 40 Wistar albino species rats with weights ranging from 250-400g were randomized into 5 groups.Control, ulcerative colitis (TNBS), ulcerative colitis + betanin (Bet), ulcerative colitis + mesalazine (Mes), ulcerative colitis + mesalazine + betanin (M+B) groups were formed.37% ethanol via intragastric and normal saline via intrarektal to the control group; TNBS to ulcerative colitis; right 6 hours after the experimental colitis was created, ulcerative colitis + mesalazine group is aplied 100 mg/kg mesalazine in the first one of the 3 times; right 6 hours after the experimental colitis was created, right 6 hours after the experimental colitis was created, ulcerative colitis + betanin group is aplied 150 mg/kg betanin the first one of the 3 times, ulcerative colitis + mesalazine + betanin group is aplied 150 mg/kg betanin and 100 mg/kg mesalazine both of them in the first one of the 3 times was applied.Proinflammatory cytokines (TNF- alpha, IL 1- beta, IL-6), oxidant/antioxidant parameters (MDA, MPO, SOD inhibition, CAT, GPx) were studied from the samples taken.Tissue damage and degree of injury were examined for pathological evaluation. Findings: When the treatment groups and TNBS groups were compared, statistically significant results were found in MDA, MPO, SOD inhibition and GPx values from oxidant/antioxidant parameters (p<0.05).Statistically significant results were not found only in the CAT value. We think that this is related to the completion of our work in a short time. When the treatment groups and TNBS groups were compared, TNF- alpha, IL 1- beta, IL-6 levels were found to be statistically significant (p<0.05).Although there was an observational difference in the group where betanin was used alone compared to the TNBS group, there was no statistically significant difference (p>0.05).In our study, it was found that mucosal loss and cellular inflammation levels were statistically significant in the group where mesalazine and betanin were used together in pathological evaluation compared to the TNBS group (p<0.05). Conclusion:In our study, biochemical and histological results have shown positive results in the rats that produce ulcerative colitis of betanin.The use of betanin alone or in combination with mesalazinein ulcerative colitisis promising in terms of treatment for the future.
