Abstract (EN)
In this master's thesis study, it is used in anorexia, cholesterol, and diabetes by local people that grows naturally in the Eastern Anatolia region, and until today hasn't been the isolation of bioactive compounds and various activity studies, it is aimed to determine the isolation of bioactive compounds with such chromatographic methods. C. bulbosum was extracted under room conditions with solvents of increasing polarity, including n-hexane, chloroform, acetone, methanol, and water, and a total of 5 different extracts were obtained. The antioxidant activities and anticholinesterase activities of each extract were determined. The antioxidant activities of the Chaerophyllum bulbosum were determined using five different methods (β-carotene-linoleic acid, DPPH free radical scavenging, ABTS cation radical scavenging, CUPRAC reducing power capacity, and metal chelating). In addition, the anticholinesterase activity of the plant extracts was investigated according to the Ellman method. In the light of the data obtained, extracts of C. bulbosum plant, which had high antioxidant and anticholinesterase activities, were isolated by appropriate chromatographic methods in an antioxidant activity controlled manner. From the fractions with high activity, nine compounds were purified using the methods such as Column Chromatography, Thin Layer Chromatography as well as Recycling HPLC. The structures of the purified compounds were analyzed by spectrophotometric methods such as IR, UV, NMR [1H-NMR, 13C-NMR, and 2D-NMR (COSY, HMQC, and HMBC)], mass spectrometry methods (EI-MS, and HRMS). The antioxidant and anticholinesterase activities of the pure compounds whose structures were illuminated were determined separately by the methods used above. Thus, the relationships between the activity of the purified bioactive compounds and their structures were established. With this thesis study, the structure of bioactive compounds obtained from active extracts of C. bulbosum was elucidated for the first time. The antioxidant and anticholinesterase activities of the extracts of C. bulbosum, an edible species grown in Turkey, and the pure compounds obtained were carried out in detail for the first time with this thesis study.
