A study on the effect of black seeds (Nigella sativa) oil on immune system of rainbow trout (Oncorhynchus mykiss)

In this study, changes in some non-specific defence mechanisms of rainbow trout (Oncorhynchus mykiss) exposed to aqueous extracts of Nigella sativa were investigated. A total of 900 fish (mean weight 102.30 g) were used in the study. Fish were treated with black seed oil by using 3 different application methods. At first method, the fish were fed with % 0.1, 1.0 ve 10 doses of black seed oil during 21 days period. At second method, % 0.1, 1.0 ve 10 doses of black seed oil was injected to the fish. At third method, % 0.1, 1.0 ve 10 doses of black seed oil were smeared up lateral line of fish during 3 days. After these applications, blood sampels of fish were taken at 3., 7., 14. and 21. days.Hematocrit (Ht), Leucocrit (Lt), Erythrocyte, Leucocyte, Nitroblue Tetrazolium (NBT) activate, Protein level and Total immünoglobulin levels were determined.Consequently, the highest levels of hematocrit, erythrocyte, leucocyte, nitroblue tetrazolium (NBT) were occurred at smearing method. Differences in Leucocrit levels of different groups that practiced three method were not important. The highest levels of total protein were occurred at feeding method. The highest levels of total Ig were occurred at injection method but it was not parabolic. Nonetheless smearing method had a parabolic flexuous. It was observed that nutritive characteristics of black seed oil at groups with feeding method were effective by increasing protein levels even in minimum levels.Key Words: Black seed oil, rainbow trout, Nigella sativa, Oncorhynchus mykiss, non- spesific immune parameters, immune system of fish.
