Abstract (EN)

In this study ; In our country, especially in our region, the seedlings produced fro In this study ; Detection of the presence of Citrus psorosis virus by molecular methods in the seedlings produced from "species and varieties" registered to the Subtropical Fruits Research and Application Center, which is the source of citrus cultivation, and in orchards of various ages, which were reproduced from different sources and grown in the field, and comparison of those detected in breeding stocks and those determined in the field. and to characterize the detected isolates by molecular methods between the years 2019-2021. The possibility of spreading viruses to larger production areas with new nontype isolates was therefore also investigated. In the study, samples were collected from 205 symptomatic and non-symptomatic new citrus varieties from Çukurova Region and tested molecularly. Only 6 of the isolated tNAs screened with the Cons1 and Cons2 primers had a positive response specific to the CPsV target genome with a size of 434bp. When the phylogenetic tree formed by the sequence analysis of these samples was examined, it was determined that the isolates of the Region had a high level of homology with the isolates of Spain, Tunisia and Italy. The agent was determined macroscopically and molecularly in the samples of Hernandina mandarin, ornamental lemon named Buddha's Hand and Meyer lemon, which are among the new citrus varieties produced in our region in the last 10 years.
