Abstract (EN)

The investigation of tbacterial translocation and inflammatory changes in an experimental intestinal obstruction model in splectomize ratsIntroduction and aim.Intestinal mucosal injury and acute infllammatory response, as a result of intestinal obstruction, cause the damage of intestinal barrier and bacterial translocation. It is belived that splenectomy can reduce the bacterial translocation with delaying the response of macrophage to the biomaterials. In this study, we aimed to investigate the effects of intestinal obstruction on bacterial translocation and inflammatory response in splenectomized rats.Materials and MethodsIn our study, 24 rats were used. Rats were divided into 3 groups, as follows; group 1 sham; group 2, intestinal obstruction; group 3, intestinal obstruction and splenectomy. 24 hours after the initial laparotomy, rats were sacrified with intra-cardiac blood taken and immediately underwent laparatomy. Peritoneal swap, mesenteric lymph node, liver and ileal specimens were taken in steril conditions. Samples were examined microbiologically, biochemically and histopathologically.ResultsCompared with the sham and intestinal obstruction, there were significantly differences in terms of bacterial translocation on peritoneal swap, mesenteric lymph node and liver tissue. In addition we observed significantly differences in terms of ileal mucosa damage and the inflammatory changes of ileum, mesenteric lymph nodes and liver. We observed a significant differences in terms of biochemical analysis of the inflammatory response, as follows; paroxanase, total oxidant status, tumor necrosis factor- ?, interleukin-6, interleukin-1ß, and C-reactive protein, but total antioxidant status and asymmetric dimethyl arginine were similiar when compared the groups. ,Compared with intestinal obstruction and intestinal obstruction + splenectomy groups, the microbiological, histopathological and biochemical results were similar.ConclusionIntestinal mucosal injury, as a result of increased inflammatory cyctokines and oxidative stres caused by intestinal obstruction, led to increased bacterial translocation and inflammatory changes in ileum, mesenteric lymph nodes and liver. Splenectomy had no effects on bacterial translocation and inflammatory changesKey words; intestinal obstruction, splenectomy, bacterial translocation.
