Abstract (EN)

Non-Hodgkin's Lymphoma (NHL) is the most common subtype of cell lymphoma (DLBCL) in cells that are the origin of B lymphocyte cells. In this study, mAb and mAb fragment formulations will be prepared and targeted to CD20 epitopes which are presented at the surface of B lymphocytes and show over-expression. Early and accurate diagnosis of NHL is targeted with these developed formulations. 3 different formulations will be prepared within this study, in addition to these formulations, if necessary materials and products could be provided, additional 3 different formulations will also be prepared. CD20 specific Anti-mouse mAbs and CD20 specific Anti-mouse mAb Fragments will be used in the 3 main formulations. CD20 Anti-mouse mAb and CD20 Anti-mouse mAb Fragment will be labeled with Zirconium-89 (89Zr), while only CD20 Anti-mouse mAb Fragment will be labeled with Gallium-68 (68Ga). By this way, resulting formulations will be as follows: 1. 89Zr labeled CD20 Anti-mouse mAb, 89Zr labeled CD20 Anti-mouse mAb Fragment and 68Ga labeled CD20 Anti-mouse mAb Fragment. The most effective biotechnology product will be determined by comparing the89Zr labeled CD20 Anti-mouse mAb and mAb fragments which are labeled with the same radionuclide. On the other hand, the most effective PET radionuclide will be specified by the comparison of 89Zr and 68Ga radiolabeled mAb fragments. PET images will be taken following in-vitro cell culture and in-vivo animal studies and biodistribution studies will be conducted after the preparation of optimum formulations. Currently, there isn't any study on 89Zr radionuclide in Turkey while it is a subject that recently shows increased popularity and research studies in the world. Learning 89Zr radiolabeling protocols and bringing the know-how to our country, will significantly contribute to Nuclear Medicine and Radiopharmacy Departments in Turkey.
