Abstract (EN)
Diabetes is a lifelong chronic disease characterized by hyperglycemia. Although the drugs currently used in the treatment of diabetes have hypoglissemic effects, new antidiabetic drugs are needed because of the frequency of side effects and inability to reverse complications. In traditional medicine, many plant extracts are used as traditional therapies in the treatment of diabetes. Therapeutics used in phytotherapy have been reported to contribute to the treatment of many diseases due to their antioxidant and/or anti-inflammatory effects. R&D studies on the pharmacological mechanisms of action of medicinal plants and chemical compounds synthesized as drug candidates in vitro and in vivo animal experiments and experimental animal disease models are increasing. In this thesis, the effects of broccoli seed extract (BSE, 300 mg/kg), arugula seed extract (ASE, 300 mg/kg), combinations (MIX; 300 mg/kg (BSE, 150mg/kg+ ASE, 150 mg/kg)) and imidazole ring pyrrolidine molecule (IRPM, 4 mg/kg), which have antioxidant and anti-inflammatory properties, on cognitive function in STZ-induced diabetes model in high-fat diet with caramel fed Swiss albino male mice were investigated by Barnes Maze test. Determination of antioxidant (SOD), anti-inflammatory (TNF-alpha) and apoptotic activity (BCL-2 and BAX) of the respective herbal extracts and synthetic chemicals and observation of their effects on AChE level in isolated brain tissues of mice were performed by ELISA, an in vitro experimental method in liver, kidney and brain tissues. In summary, the experimental data from the in vitro study suggest that MIX has anti-inflammatory, hypoglycemic and anti-apoptotic effects, while BTE has anti-inflammatory, anti-apoptotic and AChE activity inhibitory effects. In vivo experimental data suggest that BTE, RTE, MIX and IHPM may have therapeutic effects on cognitive functions in diabetic mice.
