Isolation and characterization of new Bacillus sp. strains producing industrial enzymes

Abstract (EN)

In this study, ?-amylase, xylanase and CMCase producing alkaliphilic, halophilic and thermophilic Bacillus sp. strains were isolated from Van Lake with alkaliphilic soil, Tuz Lake with halophilic soil and Jerusalem artichoke field of Adana.Bacillus sp. VA-24 strain of Van Lake, produces the ?-amylase with molecular weight of approximately 120 kDa, temperature and pH optimum of 90 °C and 11.0, respectively. Thermostability of the enzyme was stable up to 100 °C.Bacillus sp. strain TGB-3 of Tuz Lake produces two proteins with xylanase main activity, molecular weight of approximately 52 and 48 kDa and ?-amylase (side activity) with molecular weight approximately of 123 kDa. Halophilic xylanase has temperature and pH optimum of 60 °C and 7.0, respectively, while halophilic ?-amylase has temperature optimum and pH optimum of 50 °C and 7.0 respectively. Thermostability of xylanase was determined to be stable up to 70 °C.Bacillus sp. strain YET-1 produce thermophilic CMCase with molecular weight of approximately 48 kDa and thermostability of the CMCase were reached up to 90 °C. Optimum temperature and pH of the CMCase 50 °C and 7.0, respectively.Key Words: Bacillus, ?-amylase, CMCase, xylanase, extremophile
