ABSTRACT THE EFFECT OF DİFFERENT GRAFTİNG METHOD STOGRAFTİNG SUCCESS İN CHESTNUT (Cestanea sativa Mill.) Elif Dudu ASLAN Düzce University Graduate School of Natural and Applied Sciences, Department of Forest Engineering Master's Thesis Supervisor: Assist. Dr. Öğr. Şemsettin KULAÇ March 2019, 37 pages Chestnut (Cestane sativa Mill.)Trees in the Black Sea in Turkey, a wide distribution in the Marmara and Aegean regions. It is also locally distributed in some points in the Western Mediterranean Region. Especially, there are populations consisting of monumental individuals in Isparta-Sütçüler, Antalya-Selge and Antalya-İbradı towns. Chestnuts are distributed in a wide area in Turkey in recent years due to ink disease and cancer death and the body is exposed to too much loss. The fact that the chestnut is capable of exile helps the species to survive naturally against these diseases. Many biological, chemical or mechanical studies are being carried out to protect the species against cancer. In addition, the General Directorate of Forestry, which aims to increase the awareness of the people of the country, has some studies. Breeding of the chestnut (vaccination, selection, genetic variation, etc.) should be continued by increasing the work. This study consists of two stages. First of all, the effect of different vaccine methods (splitting, slinky, flaky eye and goat's foot) on vaccine boiling and thus on the success of vaccination were investigated. Secondly, the effect of ambient temperature on grafting success was investigated. In this context, 6 different ambient temperatures (10°C, 15°C, 20°C, 25°C, 15/5°C (16/8 hours), 20/10°C (16/8 hours) constant and variable temperatures and tunnel sera). In this study, one-year-old tubed seedlings grown from chestnuts collected from natural forests in Akçakoca Kestane Bayırı region were used. In this study, Sinop-Erfelek origin vaccines were used. Tunnel greenhouses belonging to Düzce University Forestry Faculty were used for the study. As tube mortar 30% forest soil, 30% peat sand, 30% river sand and 10% organic sheep manure were used. In the study, germinated and germinated seeds were first planted in polyethylene tubes in chestnut seeds germination cabinets. One-year old seedlings were vaccinated on 07.03.2015. For the first part of the study, vaccines were expected to boil in the tunnel greenhouse environment. For the second part, the success of boiling in different germination cabinets in Seed Technologies laboratory was investigated. On 30.06.2015, the vaccines were checked and counted. In conclusion, when the effect of the vaccine method on the retention success was examined first, the highest retention success was obtained from yongalıgöz with a ratio of 63.33%. Vaccines have a second success rate of 54.44%. The lowest retention success was with the Keçiayağı method with 12.75%. In the yarmaaşı method, a retention success of 43.33% was achieved. Considering the effect of different ambient temperatures on the success of vaccination, the highest retention success was obtained at a variable temperature of 15/5°C (16/8 hours) with a rate of 81.56%. The retention performance of the vaccines at a constant temperature of 15°C was the second with a ratio of 78.3%. The lowest retention success was at a constant temperature of 18,66% to 25°C. The second worst performance was at 33.33% at a constant temperature of 10°C. Retention achievements at other temperatures were found to be 20/10°C (16/8 hours), 20°C, and tunnel greenhouse environments, respectively 50%, 60% and 54.43%.
