Abstract (EN)
Gene expression studies in recent years have drawn attention to strikingly altered miRNA expressions in cancer cells. miRNAs can show a tumor suppressor or oncogene feature depending on the character of the gene they target. It is known that mir-105, a miRNA, also functions as a tumor suppressor or oncogene, depending on the cancer type. In our study, in order to examine the effect of miR-105-5p in colon and hepatocellular carcinoma (HCC); HCT116 and Huh7 cells were transfected with miR-105-5p-mimic, inhibitor and negative control. Transfection of miR-105-5p; In HCT116 and Huh7 cell lines; Its effects on cell proliferation, colony formation, invasion, hTERT, p21 and p53 expression were investigated. It was observed that downregulation of miR-105 reduces colonization in HCC, while upregulation reduces colonization in HCT116 and expression of hTERT, p53, p21 genes. However, downregulation of miR-105-5p in Huh7 cells decreased the expression of hTERT and p53 In addition, downregulation of miR-105-5p increased the proliferation of both cell lines. Finally, increased expression of miR-105-5p increased colonization in both Huh7 and HCT116 cells. In addition, inhibition of miR-105-5p resulted in increased proliferation in both cell lines. In October, the expression of miR-105-5p and let-7c genes were checked in transfected Huh7 and HCT116 cells to check whether the transfection process had occurred, but no results were obtained.
