Abstract (EN)
Hidradenitis suppurativa is a chronic inflammatory skin disease. To the best of our knowledge, the expression of trace amine-associated receptor 1 and its relationship with the janus kinase/signal transducer and activator of transcription pathway have not previously been evaluated in inflammatory skin diseases. In our study, lesional and perilesional skin samples obtained from patients with hidradenitis suppurativa and skin samples obtained from healthy individuals were compared. The gene expression levels of trace amine associated receptor 1, interleukin-6, interleukin-6 receptor, Janus kinase 1-3, signal transducer and activator of transcription 1-6, and suppressor of cytokine signaling 1-3 were measured by the real-time polymerase chain reaction method. The protein levels of interleukin-6, trace amine associated receptor 1, signal transducer and activator of transcription 3, and phosphorylated signal transducer and activator of transcription 3 were evaluated by Western blot analysis in samples prepared by combining three tissue samples in one pool. In the skin samples of patients with hidradenitis suppurativa, the gene expression levels of interleukin-6, interleukin-6 receptor, trace amine associated receptor 1, janus kinase 1-2, signal transducer and activator of transcription 1, 3-5, and suppressor of cytokine signaling 1-3 were found to be higher compared to the control group. In Western blot analysis, the protein levels of trace amine associated receptor 1, signal transducer and activator of transcription 3, and interleukin-6 were increased compared to the controls. No significant difference was observed in phosphorylated signal transducer and activator of transcription 3 protein levels. Our study reveals the expression of trace amine associated receptor 1 as a potential target molecule that may play a role in the pathophysiology of hidradenitis suppurativa and evaluates its relationship with the Janus kinase/signal transducer and activator of transcription signaling pathway. It also demonstrates that suppressor of cytokine signaling 1-3 makes an important contribution to the regulation of inflammation. The data obtained will contribute to further research aimed at the development of new therapeutic agents for diseases such as hidradenitis suppurativa.
