Abstract (EN)

ABSTRACT EXAMINATION OF THE EFFECT OF UROMODULIN (TAMM-HORSFALL PROTEIN) IN URINE ON ESCHERİCHIA COLI, KLEBSIELLA PNEUMONIAE, PSEUDOMONAS AERUGINOSA, ENTEROCOCCUS FAECALIS AND CANDIDA ALBICANS Urinary tract infections (UTIs) are one of the most common bacterial infections, affecting 150 million people worldwide each year. The most common microorganism causing UTI is uropathogenic Escherichia coli (UPEC). Microorganisms such as Klebsiella pneumoniae, Pseudomonas aeroginosa, Enterococcus faecalis and Candida albicans are also seen as causative agents. Uromodulin (UMOD) is the most abundant protein in the urine, known to be involved in the innate immune mechanisms of the urinary system. The aim of this study is to show that uromodulin in the urine is a molecule that prevents the formation of UTI or contributes to its treatment, and to show that this molecule can be used as a support in such infections in the future. A total of 35 healthy volunteers, 18 female and 17 male, aged between 22 and 72 years were included in the study. The above-mentioned microorganisms, which are known to be the most common UTI agents and whose amounts are known in cfu/ml, were inoculated into individual urine samples of each healthy volunteer and their amounts were measured in cfu/ml after 24 hours of incubation. Then, by measuring the amount of UMOD in the urine samples cultivated and uncultivated microorganisms; It has been shown how the UMOD levels in the urine have an effect on removing these microorganisms from the urine or stopping their reproduction, and how these microorganisms affect the UMOD level. IBM SPSS Statistics Version 22.0 package program was used in the statistical analysis of the data, and the result was considered statistically significant in all analyzes (p<0.05). E. coli, K. pneumoniae, P. aeruginosa, E. faecalis and C. albicans; A statistically significant decrease was found when the UMOD amounts measured after incubation and before incubation were compared. These results show that uromodulin in the urine affects the amount of uropathogen with a bacteriostatic and/or bactericidal effect; We think that more patient numbers or new multicenter studies are needed on this subject.
